Objective Investigate the early metabolic characterization in plasma of rats exposed to whole thorax irradiation (WTI) in order to disclose the early biomarkers of non-uniform local irradiation and provide the experimental evidence of the mechanism of radiation injury. Methods Male Wistar rats were randomly divided into healthy control group and irradiation group. The dynamic alternations of metabolites in plasma of rats on the day 2, 3 and 5 after WTI were detected by gas chromatography mass spectrometry (GC-MS), and the differential metabolic features and metabolic pathway concerning radiation injury were analyzed. Results Principal Component Analysis (PCA) showed that the metabolic differences between control and WTI groups became more obvious along with the time after irradiation. Compared to the control, some metabolites such as inosine, alanine, palmitic acid, stearic acid, lactic acid and glucose were significantly increased during 3-5 days after WTI(F=13.51, 5.20, 9.87, 10.59, 7.05, P<0.05), while some metabolites such as methionine, valine, threonic acid, fumaric acid, malic acid, α-ketoglutaric acid, phosphoric acid and lysine were obviously decreased(F=15.68, 5.63, 3.78, 12.83, 2.88, 8.93, 4.68, 6.43,P<0.05). Conclusions The metabolic profile changes in rat plasma after WTI mainly include amino acids metabolism, tricarboxylic acid (TCA) cycle and lipid metabolism. |
