Objective To compare the difference of 3'-deoxy-3'-18F-fluorothymidine (18F-FLT) and 2'-deoxy-2'-18F-fluoro-D-glucose (18F-FDG) in the ultra-early biological response of esophageal carcinoma cells after irradiation. Methods After 2, 4 and 8 h of irradiation with different doses (5, 10, 15 Gy), the uptake ratio of 18F-FDG and 18F-FLT, survival, and ATP level of esophageal carcinoma cells were detected. Results After 2 and 4 h of 5 Gy irradiation, the cellular 18F-FDG uptake ratio was decreased by 9.45% and 16.4%. After 2 h of 15 Gy irradiation, this uptake ratio was increased by 26.5% (t=3.04, P<0.05). But the 18F-FDG uptake ratios of the other experimental groups were decreased remarkably (F=25.75, P<0.05). On the other side, after 2 h of 5 Gy irradiation, the 18F-FLT uptake ratio had no statistically difference [(3.65±0.17)% vs (4.00±0.17)%, P>0.05], and 18F-FLT uptake ratios of the other groups were decreased notably (F=33.93, P<0.05). The relative survival of esophageal carcinoma cells after 2, 4 and 8 h of irradiation with different doses (5, 10, 15 Gy) has no significant difference. Line regression analysis indicated a significant correlation between 18F-FLT and ATP levels (r=0.887, P<0.05), whereas a less correlation was found between 18F-FDG and ATP levels (r=0.622, P>0.05). Conclusions Both 18F-FDG and 18F-FLT can evaluate the ultra-early irradiation response of esophageal carcinoma cells, but 18F-FLT may be more specific than 18F-FDG in monitoring the ultra-early irradiation response. |
