Objective To establish and identify the radiobiological characters of VEGF-transfected cell line. Methods The expression vectors of VEGF165 sense and antisense gene were transfected into vascular endothelial cells. The stable cell line was established and its radiobiological effects were evaluated using MTT method and micronucleus assay. Results The sense and anti-sense expression vectors were successfully transfected into the vascular endothelial cells and the stable cell lines were established. The cell line transfected with sense expression vector of VEGF165 gene showed enhanced proliferation (10.8±1.9)%(P<0.05). The micronucleus frequency(MNF) was reduced from (53.5±6.61)% to (42.5±2.46)%(P<0.01),and the micronucleus cell frequency(MNCF) reduced from (31.6±5.23)% to (23.7±2.27)%(P<0.05). The opposite results were obtained in the anti-sense transfected cell line, which showed the reduction in proliferation(-9.2±2.8)%(P<0.05),but the increase in MNF(62.1±1.96)%(P<0.01) and MNCF (38.1±1.47)%(P<0.05). Conclusions VEGF has a protective effect on the vascular endothelial cells exposed to γ ray irradiation, which may illustrate a new evidence of therapy for radiation injury. |
