Objective To explore the inhibitory effect and radiation sensitization of recombinant adenovirus encoding human p53 tumor suppressor gene (rAd-p53) on human lymphoma cell lines. Methods Human lymphoma cell lines Raji and Daudi were treated with rAd-p53, radiation therapy and combined treatment,respectively. The cell growth inhibition was assessed by MTT. The p53 protein expression was detected by Western blotting, and p53 mRNA was detected by RT-PCR. Results The MTT results showed that the inhibitory effect and radiosensitivity enhancement of rAd-p53 on human lymphoma cell lines were not obvious [Raji:(27.5±4.1)%;Daudi:(28.1±1.6)%]. The results of Western blotting and RT-PCR showed that extrinsic p53 protein and p53 mRNA were expressed to some degree, but not at high-level. In addition, the results didn't demonstrate obvious radiosensitivity enhancement. Conclusions The role of inhibition and radiosensitivity enhancement of rAd-p53 was not significant on human lymphoma cell lines. |
