Objective To explore the changes in IFN-γ secretion in intestinal intraepithelial lymphocytes (IEL) after radiation with 60Co γ-ray of 8 Gy and its influence on the proliferation of IEC-6. MethodsELISPOT was used to detect the changes of IFN-γ secretion in IEL after radiation. The influence of IEL on the proliferation of crypt cell line from rat jejunal epithelium (IEC-6) was analyzed by MTT assay, using the cell models of IEC-6 and incorporate_ and double_cell culture system.Results IFN-γ secretion in IEL after radiation was increased. And the culture supernatant of IEL after radiation had inhibitory action on the proliferation of IEC-6. The double-cell culture system could decrease this inhibitory action, and adding anti-IFN-γ have the same effect.Conclusions IFN-γ secretion in intestinal intraepithelial lymphocytes is enhanced after radiation and it has the inhibitory action on the the proliferation of IEC-6. Cell to cell contact may be an important way for IEL to inhibit the proliferation of IEC-6. |
