Abstract::
      Objective To compare the course of radiation pulmonary injury in C57BL/6J and C3H/HeN mice.Methods C57BL/6J and C3H/HeN mice irradiated with 20 Gy 60Co gamma rays were used as animal models. Sirius red staining and hydroxyproline measurement were used to detect the distribution of type Ⅰ and Ⅲ collagens and the content of pulmonary hydroxyproline. Immunohistochemistry was used to observe the changes of the expressions of fibronectin (FN) and laminin (LN) and alpha-smooth muscle actin(α-SMA) in the lung.Results The changes in the lungs in C57BL/6J mice were as follows: the pathological changes undergone interstitial pneumonia, proliferation and fibrosis; significant increase of collagen deposition; FN increased rapidly to a significantly higher level at 1 and 3 months after irradiation than that in the control (P<0.01), and then decreased gradually to a normal level at 6 month after irradiation; LN elevated gradually after irradiation; the expressions of α-SMA were more intense than that in C3H/HeN mice. The changes in the lungs in C3H/HeN mice were as follows: the pathological changes mainly developed institial pneumonitis; no significant changes of FN expression was observed after irradiation compared to that in the control; LN increased significantly at 1 and 3 months after irradiation, and then decreased gradually.Conclusions The models of radiation pulmonary fibrosis_sensitive and_resistant were established by irradiation with gamma rays, C57BL/6J mice developed late radiation pulmonary fibrosis, and were characterized by the significant accumulation of collagen. C3H/HeN mice did not develop radiation pulmonary fibrosis.
