Objective To explore the genomic instability induced by 60Coγ rays. Methods The cloning efficiency and micronucleus efficiency of normal human liver cell irradiated by 60Co γ rays were detected, and the method of single cell gel electrophoresis (SCGE) was carried out to measure DNA chains damage. The fast-growing cells were divided into different dose-groups and then irradiated by 60Coγ rays. After 40 populations doubling, the progenies were secondly irradiated with 2 Gy 60Coγ rays. Results The cloning efficiency decreased with the increase of doses after the initial irradiation. After the survival cells were given second irradiation, both results of SCGE and micronucleus frequency showed that the second damage was correlated with the original irradiation doses. Conclusions 60Coγ rays can not only induce the immediate biological effects in liver cells, but also lead to the genomic instability in the descendants that leads to an enhanced frequency of genetic changes occurring among the progeny of the original irradiated cell. The expanding effect of second event helps to study the genomic instability. |
