Abstract::
      Objective To study the effect of WBI with X rays on the expression of LAMP-1 in the splenocytes of mice. Methods The LAMP-1 positive cells from mouse splenocytes were detected with flow cytometry (FCM) at different times(0,2,4,8,16,24 and 48 h) after WBI with X-rays of 0\^075 Gy and 2.0 Gy and after WBI added by Con A stimulation. Results In view of time-course effect,the LAMP-1 expression in the splenocytes was significantly inhibited at 8,16, 24 h after WBI by 2.0 Gy X-rays while the LAMP-1 expression was markedly enhanced at 2 h and suppressed at 48 h after WBI by 0.075 Gy X-rays. Compared with control group,the LAMP-1 expression in the splenocytes was significantly up-regulated with Con A stimulation(10 μg/μl) after WBI with 0.075 and 2.0 Gy X rays. With Con A added, the effect of 2.0 Gy X rays was greater than that of 0.075 Gy. Conclusions LAMP-1 plays a role in the transduction of immune signals induced by ionizing radiation and is closely correlated with the activity of immune cells. The high dose irradiation inhibits the LAMP-1 expression while the low dose likely causes the opposite biological effect in the early time. Con A enhances the expression of LAMP-1 on the surface of immune cells.
