Objective To observe the formation of acini structure of prostate cells under 3-dimensional (3_D) condition and the effects of radiation on it. Methods RWPE-1 cells were cultured under 3-D condition, Western blot was used to detect the expressions of Beclin 1,p53 and p21,in Beclin 1 knock-down cell model,i.e.,RWPE-Bi, immunofluorescence staining and confocal microscopy were used to detect the morphological characteristics of RWPE cells irradiated by 60Co. Results In 3_D culture,acini was observed,characterized by sphroid structure and cell polarity.After radiation,the acini was destructed and cell death was found,especially in RWPE-Bi compared with RWPE-1. Conclusions Radiation could suppress the formation of acini structure of prostate cells and Beclin 1 might play a role in this process. |
