Objective To investigate the protective effects of platelet factor 4(PF4) on bone marrow stromal cells (BMSCs)in the irradiated mice and explore its mechanism. Methods Thirty male mice were randomly divided into three groups; mice in one of the three groups were exposed to whole-body irradiation of 5.0 Gy 60Co gamma rays. The mice were intraperitoneally injected with PF4 (50 μg/kg) at 26 h and 20 h before the irradiation. The CFU-F was counted at 3 d,7 d, and 14 d after the cell culture, respectively. Cell cycle of BMSCs cultured in vitro after irradiation, were analyzed by FACS at 10 d since the culture. Results Compared with the group protected by PF4,the difference of number of CFU-F at 3 d in irradiation group was not statistically significant. But the difference at 7 d or 14 d was statistically significant. The cell percentage of G0+G1 phase in the irradiation group increased, while the percentage of S phase and G2+M phase decreased. Conclusion PF4 could protect BMSC in the irradiated mice and promote recovery of hematopoiesis. |
