Objective To provide basis for clinical application of cord blood. Methods Using freshly isolated mononuclear cells without or with 62 mGy low dose irradiation by 60Co γ-rays and with 3H-TdR incorporation,release assay,ELISA,flow cytometry,MTT and CFU-GM were performed to study the quantity of cord blood,the number of MNC,active MNC percentage,activities of T cells and NK cell,the expressions of membrane molecules of T lymphocytes,levels of IL-2 TNFα,sIL-2R,cytotoxity of LAK cells aganst target cells(K562,HL-60),CFU-GM,activity changes of T cells and NK cells after irradiation by low dose γ-rays,activity of colony-stimulating factors,respectively. Results The quantity of cord blood was ranged 100-150 ml,and there was no relation to the MNC number.The cord blood was rich in hematopoietic stem /progenitor cells.Immunological activities of T and NK cells was low,but they were enhanced after low dose γ-ray irradiation.The serum of cord blood was rich in colony-stimulating factors,chiefly IL-3.The clinical application of cord blood for treatment of leukopenia would be obviously effective. Conclusion The prospect for clinical application of human cord blood is encouraging. |
