Objective To study the relationship between the proliferation of Deinococcus radiodurans (Dr) and the concentrations of superoxide anion and hydrogen peroxide produced in irradiated Dr with different doses. Methods The release of O2. from Dr was determined with cytochrome C reduction method. Then diphenyleneiodonium (DPI), the inhibitor of NADPH oxidase, was added into the culture medium. The concentration of H2O2 in Dr was measured with a laser confocal scanning microscope (LCSM), and the release of O2. and Dr’s proliferation in the same circumstance with 3H-TdR incorporation method. Results The proliferation of Dr was related with the concentrations of O2. and H2O2. The radiation dose less than 1 kGy could stimulate proliferation of Dr, Whereas the proliferation was suppressed by higher dose of radiation. Conclusion Certain concentrations of O2.,H2O2 are essential to the proliferation of Deinococcus radioduras. |
