Abstract::
      Objective To investigate the effects of γ rays on the transcriptional activity of SP1, AP1 and Smad3-Smad4 in pulmonary fibroblasts. Methods Rat pulmonary fibroblasts were separated,cultured and then irradiated with 3, 5 and 8 Gy 60Coγ-rays. The transcriptional activity of SP1, AP1 and Smad3-Smad4 and the expression of PAI-Ⅰ and type Ⅰcollagen were observed by means of EMSA, immunoblotting and immunohistochemistry. Results In normal fibroblasts, AP1, Smad3-Smad4 formed weak specific shift binding blot,but SP1 did not. After irradiated with 3, 5 and 8 Gy 60Coγ-rays, the transcriptional factors AP1, SP1 and Smad3-Smad4 were activated and the expression levels of type Ⅰcollagen and PAI-Ⅰ were increased. Conclusion A certain dose of γ-rays may promote the activation of transcriptional factors SP1, AP1 and Smad3-Smad4 and thus increase the expressions of PAI-Ⅰ and type Ⅰ collagen.
