| Objective To resume the ability of revascularization of the irradiated tissue by VEGF gene transfection. Methods Each one of the mice was irradiated for 2 Gy every time and 30 Gy in total on the right leg. The irradiated muscular tissues of animal models were transfected with 200 μg pcDNA3/VEGF165 per animal on the right leg and another 200 μg two weeks later. After the gene therapy we determined VEGF165 mRNA,VEGF protein, vessel density, average diameter of vessels and average sectional aera. We also observed ultrastructure of the vessels at the same time. Results VEGF165 mRNA and VEGF protein were highly expressed in the irradiated tissue. Vessel density, average diameter and average sectional aera were also not different from those on the left leg, and the configuration and ultrastructure of vessels returned to normal. Conclusion VEGF gene therapy can resume the ability of revascularization of irradiated tissue, which provides the bases for its clinical application. |
