| Objective To investigate the effect of whole body X-irradiation (WBI) on metallothionein-1 (MT-1) gene transcription level in mouse tissues and its dose-effect relationship. Methods MT-1 mRNA levels in mouse liver,brain,thymus and spleen were measured using Northern blot assay. Results Time-course experiment showed that MT-1 mRNA levels in mouse liver and thymus gradually increased after WBI,reached a peak at 4 h after WBI and recovered normal level at 24 h. MT-1 mRNA levels in mouse liver and thymus increased with increase of irradiation dose at 4 h after 0.5~6 Gy X-ray WBI. But there was no obvious change of MT-1 mRNA levels in mouse brain and spleen. Conclusion MT-1 mRNA levels in mouse liver and thymus significantly increase after WBI and showed a dose-dependent trend. But there is no obvious change in MT-1 mRNA levels in mouse brain and spleen. These Results indicate that increase of MT-1 mRNA in mouse tissues after WBI is tissue-specific,and MT protein synthesis is regulated through MT-1 gene transcriptional elevation. |
