| Objective To investigate HR24L repair complex after DNA damage induced by radiation and provide biochemical evidence of the participation of HR24L in DNA excision repair and recombination repair. Methods Cotransfection and coimmunoprecipitations were employed to detect the presence of key repair proteins in HR24L compex. Results XPA was present in HR24L precipitations 2 h after 20 J/m2 UVirradiation.When cells were irradiated with 20 Gy γ-rays,Ku70 was precipitated with HR24L after 2 h and Rad52 was found in HR24L complex after 8 h. Conclusion By showing direct biochemical evidence for the presence of excision and recombination proteins in HR24L complex after radiation damage,the argument that HR24L participates in multiple repair pathway is reinforced. |
