Abstract::
      Objective To investigate the role of ceramide and JNK signaling pathway in UVA-induced apoptosis. Methods Cell death rate and apoptosis were examined by MTT assay and DNA ladder analysis respectively.Activation of JNK signaling pathway was analyzed by Western blot. Results UVA-induced apoptosis was markedly observed in lymphocyte JY,but not in MS 1418 cells from Niemann-Pick type D patients with an inherited deficiency of acid sphingomyelinase,resulting in inhibition of ceramide production.Once exogenous ceramide was administered,apoptosis occurred in both JY and MS 1418 cells,indicating UVA induced apoptosis via ceramide.UVA radiation activated JNK signaling pathway in JY but not in MS 1418, and exogenous ceramide stimulated JNK signaling pathway in both cells.The results indicated that ceramide was on the top of JNK signaling pathway. Conclusion UVA-induced apoptosis depends on ceramide and JNK signaling pathway.
