Objective To study the effects of brachytherapy on the gene expessions of elastin and elastase in cultured rat vascular smooth muscle cells (VSMCs). Methods Rat VSMCs cultured in DMEM containing 10% FBS were irradiated by60 Co γ-rays at 0, 7, 14, 28 Gy respectively. Then mRNA levels of elastin and elastase were determined by reverse transcription competitive PCR(RT-PCR). Results Brachytherapy inhibited the expressions of elastase. Elastase mRNA decreased from 82.52±6.50 to 64.25±3.81 and 42.17±3.16 in VSMC irradiated with 14, 25 Gy, respectively(P <0.05). The elastin mRNA level increased from 95.17±6.41 to 72.05±5.82 and 49.77±5.62 in VSMC irradiated with 14, 25 Gy, respectively(P <0.05). Conclusion Brachytheapy inhabits the expressions of elastase and increased elastin in VSMC cells. |
