Objective To study the effect of radiotherapy on hprt gene in peripheral blood lymphocytes,and explore the possibility of radiotherapy-induced genetic damage detected by the frequency of hprt locus mutation. Methods A simple,rapid and sensitive poly-nuclear cell assay was used to investigate the frequency of hprt locus mutation in malignant tumor patients prior to and after receiving radiotherapy with a cumulative dose of 54-72 Gy. Results The results showed that radiotherapy could induce hprt locus mutation in malignant tumor patients.It was found that there was a close correlation between the frequency of hprt locus mutation and the cumulative dose:that is the larger the cumulative dose,the severe the genetic damage.At the same dose,the mutation frequency of nasopharyngeal carcinoma patients was close to that of the breast cancer patients.The frequency of hprt in malignant tumor patients was higher than that of normal,healthy people. Conclusion The frequency of hprt locus mutation may be expected to be used as a potential biological indicator for genetic damage induced by radiotherapy. |
