Abstract::
      Objective To analyse the changes of VEGF expression and apoptosis in murine bone marrow stromal cells(BMSCs) after in vivo exposure to γ-radiation. Methods Kunming strain mice irradiated with 6.0 Gy 60Coγ-rays were used to analyse the VEGF expression in BMSCs on day 7 and 14 after irradiation.Cultured BMSCs were used to extract total RNA and protein.The methods of RT-PCR and Western blotting were used to measure the relative expression levels.The apoptosis rate of BMSCs were tested by flow cytometry(FCM). Results The expressions of VEGF in BMSCs at mRNA and protein levels were all reduced significantly after irradiation and increased slowly with time.The apoptosis rate of BMSCs increased significantly after irradiation and decreased slowly with time.These values were still abnormal on day 14. Conclusion Gamma radiation inhibits VEGF expression in BMSCs and induces BMSCs apoptosis,suggesting that abnormal expression of VEGF in BMSCs is involved in the injury of hematopoietic microenvironment induced by γ-radiation.
