Objective To study the rescue effects of peripheral blood stem cell (PBSC) mobilized by anti-CD49d monoclonal antibody (McAb) combined with recombinant human granulocyte colony-stimulating factor (rhG-CSF) on lethally irradiated mice. Methods BALB/c mice received 8.5 Gy total body irradiation were transplanted with either NS (control group),PBSCs from rhG-CSF-mobilized mice (experimental group 1),PBSCs from anti-CD49d McAb-mobilized mice (experimental group 2),or PBSCs from anti-CD49d McAb and rhG-CSF-combinatively mobilized mice (experimental group 3).The white blood cells count,4-week survival rate,and numbers of bone marrow nucleated cells (BMNC),granulocyte-macrophage colony-forming unit (CFU-GM),colony-forming unit-spleen (CFU-S) were observed. Results The survival rate and WBC,BMNC,CFU-GM,CFU-S counts were all significantly higher in experimental group 3 than those in the control group,experimental group 1,and experimental group 2 (P<0.01 orP<0.05). Conclusions Anti-CD49d McAb and rhG-CSF may synergetically mobilize PBSC in mice.PBSCs mobilized by combination of anti-CD49d McAb and rhG-CSF could effectively rescue lethally irradiated mice. |
