Abstract::
      Objective In order to study the radiosensitivity and the regressive effect of cell survival curves in Suzhou human glioma cell-44, clone forming assay (CFA), 3H-TdR incorporation and MTT assay have been applied in the experiment. Methods We established irradiation groups (n=6×14) and control groups (n=12×1).The irradiation groups were divided into 14 subgroups, 6 parallel samples each, irradiated by 60Coγ-rays (2.0 Gy/min) with doses of 0.5, 1, 2, 3, 4, 5, 6, 7, 8, 9, 10, 11, 12, 13 Gy respectively.Each dose group was repeated three times. Results The radiosensitivity values SF2, D-2 were 86.96% and 17.85 Gy.The other values D0 3.20 Gy, Dq 2.35 Gy and α/β 16.98 Gy were also obtained.The cell survival curve showed a small shoulder in small dose range, and a not steep gradient(1/D0=0.32).At large doses (>10 Gy) the survival fraction declined rapidly.The sensitivity of 3H-TdR incorporation assays was 90.0%, but specificity was 88.5%.The false-positive and false-negative rates were 100% or so, while the specificity of MTT assay was lower (75.0%), and the false-positive was higher (25.0%). Conclusion Radioresistance appears in SHG-44 cells and the cell survival curve by CFA is the excellent among the three experimental assays (Q=0.007, F=4.56, PP<0.001).
