Objective To explore the possibility of radiation-induced DNA double-strand breaks (DSBs) detected by neutral comet assay as a predictor of intrinsic radiosensitivity. Method The relationship between intrinsic radiosensitivity of tumor cells and radiation-induced initial DNA DSBs, DSB repair in three tumor cell lines:K562, LS-T-117 and C6 were investigated by the neutral comet assay. Result It was found that there was a close correlation between initial DSBs and the delivered dose:that is the larger the dose, the severer the DSB.For same single dose, the different initial DSBs and intrinsic radiosensitivity measured by means of a clonogenic assay were ranked in the following descending order:K562>LS-T-117>C6, while the capacity of DBS repair:C6>LS-T-117>K562. Conclusion The results suggest both radiation-induced initial DSBs and DSB repair correlate with intrinsic radiosensitivity to a certain extent, which can be used as potential predictors of intrinsic radiosensitivity for tumor cells. |
