Objective To observe the antitumor effect of pEgr-ssEndostatin gene-radiotherapy to murine melanoma and the expression of Endostatin gene in tumor induced by radiation. Methods pEgr-ssEndostatin plasmids were injected locally into the tumor in B16 mice, and 42 h later, the tumors of half of the mice were given 20 Gy X-irradiation.The tumor growth at different times and Endostatin expression 2 days after irradiation were detected. Results Endostatin gene-radiotherapy could enhance the curative effect of radiotherapy.Both pEgr-ssEndostatin group and pEgr-ssEndostatin+20 Gy group had Endostatin mRNA expression, but the mRNA level of pEgr-ssEndostatin+20 Gy group was higher than that of pEgr-ssEndostatin group. Conclusion The antitumor effect of pEgr-ssEndostatin gene-radiotherapy is better than that of radiotherapy and gene therapy only.And Endostatin expression in tumor is enhanced after irradiation. |
