Objective For elucidating the molecular mechanism of G1 arrest induced by ionizing radiation, the transcription of p53, waf1 and gadd45 genes in irradiated bone marrow cells of mice was studied. Methods Northern blot and semi-quantitative RT-PCR were used to detect p53, waf1 and gadd45 at mRNA level. Results The results showed the p53 expression level elevated and waf1 was also up-regulated beginning from 2 h after irradiation and reached its peak at 4 h.As for gadd45, a transient increase was showed at 2 h, then decreased until 72 h.Dose-effect results showed that p53 increased 12 h after whole-body irradiation with 0 .5-6. 0 Gy irradiation, but was not in a significant dose-dependent manner.waf1 showed a dose-dependent increase, 6. 0 Gy might elevate waf1 3.41 times of that in sham-irradiatied mice gadd45 had no significant changes. Conclusion The results suggested that p53 and waf1 other than gadd45 might play important roles in G1 arrest induced by high dose radiation. |
