Objective To investigate the dose effect relationship of nitric oxide in inhibiting proliferation of pulmonary fibroblasts. Methods MTT colorimetry and immunohistochemistry were used to observe the proliferative activity and expression of ET.TGFβ.AⅡ in fibroblasts. Results Low dose 60 Co γ-ray promoted proliferation of cultured HELF cells especially 5 Gy.NO provider sodium nitroprusside released NO linearly within 30 hours by measuring the concentration of NO in the culture medium.The average increase amplitude was 4.25 μmol/L.The optimum concertration to depress hyperplasia of fibroblast was 4~5 μmol/L.The ET、TGFmRNA transcription and protein expression of ET、TGF.AⅡ were increased after irradiation. Conclusion NO can inhibit excessive proliferation and secretion of fibroblasts in a dose dependent way. |
