Abstract::
      Objective To study the delayed effect of radiation on the gene expression of anion exchanger (band 3 protein) in erythrocyte membrane. Methods Two-step liquid culture of erythroid cell was conducted by isolating blasts freshly from patients with bone marrow form of acute radiation sickness (ARS) at the 10 th year after exposure to radiation and the band 3 gene expression in the culture cells was observed using B3 mRNA in situ hybridization.The expression level of band 3 gene in total RNA from peripheral blood mononuclear cells of ARS patients was analyzed by semi-quantitative reverse transcriptase-polymerase chain reaction (RT-PCR).β-actin was used as the internal reference. Results Weak and uneven location of band 3 mRNA positive stain in the cultured late erythroblasts was shown by using in situ hybridization.The expression level of band 3 mRNA in half of the ARS patients was decreased. Conclusion The results suggest that the delayed effect of radiation might be exerted on band 3 mRNA transcription, resulting in lowered level of the transcription product and its weakened function, e.g.the decreased anion transport of anion exchanger, and prolonged course of erythropoiesis.
