Abstract::
      Objective To Compare the proliferaitve capacity,apoptosis and cell cycle alterations of human ovarian cancer cell line A2780 and its cisplatin-resistant subline A2780-CP in vitro after 60Coγ irradiation. Methods The effects of in vitro proliferative capacity of both A2780 and A2780 CP cell lines irradiated with 1~ 10 Gy gamma-rays were evaluated with microculture tetrazolium(MTT) colourmetry.The apoptosis and cell cycle of the above-mentioned cell lines were analyzed with flow cytometry. Results (1)60 Coγ-irradiation inhibited the growth of A2780 cells significantly and induced their apoptosis in a dose-dependent manner,while A2780-CP cells exhibited higher radioresistance.(2) The G1 phase cells arrested progressively and the ratio of Sphase cell decreased in A2780 cells,while the ratio of G1 phase cells and of Sphase cells increased in A2780-CP cells after exposure to 6 Gy 60Coγ-irradiation in a time-dependent manner. Conclusion The cisplatin-resistant cell line of ovarian cancer is significantly radioresistant and exhibits characteristic alterations in cells cycle after irradiation.Radiation induced apoptosis and cell cycle alterations revealed by flow cytometry can foretell the radiosensitivity of human malignant tumour cells in vitro.
