Abstract::
      Objective To investigate the possible mechanism of the effects of radiation on Bcl-xLand Bax gene transcription in 8226 and Raji cell lines with different p53 statutes. Methods Apoptosis was studied by Annexin-V-PIand flow cytometry.Bcl-xL,Bax gene transcription and the cell apoptosis induced by 60Co γ-radiation were analysed by RT-PCR and DNA released assay,respectively.The proliferation suppression of cells were studied by MTT assay. Results The transcriptions of Bcl-xLand Bax in 8226 and Raji cell lines increased abruptly and reached the their highest points-4-8h after γ-irradiation and then dropped down to the normal levels gradually.The transcriptions of Bcl-xL Bax and the ratio of Bcl-xL/Bax were correlated to apoptosis and proliferation suppression of the target cells. Conclusion The p53 gene status of the cell lines determines their sensitivity to irradiation.The 8226 cell line,having two p53 allele mutations,is more resistant to ionizing radiation than Raji with one mutation p53 allele.The apoptotic rate is correlated to the transcriptions of Bcl-xL Bax and the ratio of Bcl-xL/Bax.
