Objective To observe the protective action of IL-2,4,6,7 and IFN-γ on radiation-induced apoptosis of human tonsil lymphocyte subpopulation. Methods Human tonsil specimens were obtained from routine tonsillectomies.Lymphocytes were separated and co-cultured with IL-2,4,6,7 and IFN-γ for 24 h,then exposed to 4 Gy γ-rays.DNA gel electrophoresis and flow cytometric light scatter analysis were performed to discriminated the viable cells from dead cells,and bcl-2 expressed on it. Results The "ladder pattern" of DNA fragmentation could be seen within 4-24 h after 4 Gy irradiation.CD8+T and B cells showed higher radiosensitivity than that of CD4 + T cells.The radiation-induced apoptosis in both CD4+ and CD8+ T cells was significantly inhibited and bcl-2 expression was induced by IL-2,IL-4 and IL-7.The viability of B cells was maintained and bcl-2 was overexpressed by the presence of IL-4 but not by others in culture. Conclusion We conclude that the protective effect by each cytokine on radiation-induced apoptosis might be partly attributed to their enhancement of cellular expression of bcl-2 protein. |
