Abstract::
      Objective To study the anti-tumor effect of IFN-γ gene-radiotherapy to murine melanoma and its immunologic mechanism. Methods pcEgr-IFNγ plasmids were injected locally into tumor,and 36 hours later,the tumors were given 20 Gy X-ray irradiation.Tumor growth at different time,IFN-γ expression 3 days later and immunologic indexes 15 days later were detected. Results At 3-15 days after pcEgr-IFNγ gene-radiotherapy,the tumor growth rate was lower than that of irradiation alone group.It was also lower than that of gene thempy alone group and control plasmid combined with X-ray irradiation group significantly. Day 3 tumor IFN y expression was higher than that of plasmid treatment alone group. NK activity, IL-2 and IFN y secretion activities were higher than those of genetherapy alone and irradiation alone groups significantly. Conclusion The anti-tu-mor effect of IFN y gene-radiotherapy is better than that of either of them applied solely. Its mechanism might be conccerned with the higher expression of IFN y induced by irradiation in tumors and activation of anti-tumor immu-nologic functions.
