Objective To investigate interleukin 6 involvement in the response of multiple myeloma cell lines XG-6 and XG-7 to 60 Coγ-radiation. Methods Apoptosis was confirmed by flow cytometry and quantitated by the assay of DNA fragmentation release.The expression of Bcl-xL and Bax in XG-6 and XG-7 cells after irradiation and cultured with or without interleukin 6 was monitored by RT-PCR. Results XG-6 and XG-7 cells in the presence or absence of interleukin 6 underwent apoptosis at 24 h after γ-irradiation,but in the absence of interleukin 6 they had higher rates of γ-radiation-induced apoptosis than in the presence of interleukin 6(P<0.05) and had lower levels of Bcl-xL and Bax expression. Conclusion Interleukin 6 affects the sensitivity of the cell lines to γ-radiation-induced apoptosis and can rescue cells from apoptosis by up-regulating the expression of Bcl-xL and changing the Bcl-xL/Bax ratio. |
