Objective To explore the mechanism of decreased phagocyotic and chemotactic function of neutrophils (Neu) and the mechanism of W11-a12' s pharmacological action by studying the changes of actin of Neu in wound after total body irradiation of rats. Methods Cells were collected from incision wounds on back of rats after 6 Gy whole body-irradiation.F-actin in Neu was stained with Fluorescein-phalloidin. Results There were increased (3~12 h) and decreased (24 h, 48 h) basal levels of F-actin, a lower responsiveness to stimulation and a reduced rate of actin depolymerization after combined radiation injury compared with simple wound group, which were important causes of Neu dysfunction. Conclusion W11-a12 could improve the functional state of Neu actin, which may contribute to the improved chemotactic and phagocytotic function of Neu after administration of this drug. |
