Objective To clone ATM full length cDNA and cDNA fragments containing some functional domains and to identify proteins that interact with ATM and mediate DNA damage signal transduction in cellular response to DNA damage. Methods ATM cDNA was amplified from Marthom TM Ready cDNA kit of human leukocytes by LDPCR.ATMinteracting proteins were screened by yeast two hybrid system. Results ATM fulllength cDNA and cDNA fragments containing PI3K kinase domain, leucine zipper and proline rich region were amplified from human cDNAs.Several candidate clones that interacted with ATM PI3K domain were identified. Conclusion ATM mediates DNA damage signal transduction by interacting with many proteins. |
