Objective To assess the effects of estradiol on 60 Coγ-radiation induced apoptosis of splenic lymphocytes and thymocytes,and surface molecule expression of splenic lymphocytes. Methods Mice were whole body irradiated with 4.0 Gy γ-rays. By flow cytometry and electrophoretic analysis of DNA, the changes in apoptosis of mouse immunocytes were determined. The splenic lymphocytes were analyzed by flow cytometry with fluorescent monoclonal antibodies. Results 10 days after administration of estradiol, the characteristic DNA ladder in mice 8h after irradiation was minor than in mice without estradiol administration,indicating that the apoptotic rate reduced on flow cytometry. CD4+ T cells,CD8+ T cells and IgM+ B cells upregulated Fas,CD25 and CD69 expression,but did not so in the estradiol treated mice. Conclusion Estradiol can block CD25,CD69 and Fas overexpression,thereby inhibiting Fas mediated apoptosis inducedby γ-irradiation. |
