Objective To investigate radioresistance and cell cycle distribution of HT1080R cells after γ ray irradiation. Methods By comparison of the survival fraction between HT1080R and HT1080 using cloning assay,cell cycle distribution and gene status were investigated by FCM and PCR-SSCP,respectively. Results HT1080R was more radioresistant than its parental cell line HT1080,and after exposure to 4 Gy and 6 Gy γ-rays showed G 2 phase arrest.PCR-SSCP showed that the expression of p53 gene was normal. Conclusion G2 arrest can be found after 4 Gy and 8 Gy γ ray irradiation in HT1080R cell line,which is more radioresistant than its parental cell line HT1080,suggesting that the acquirement of radioresistance may be related to its G 2 arrest.Because of its normal gene status and expression,radioresistance of HT1080R cannot be explained by abnormal function of p53 gene. |
