Objective To explore the killing effect of retrovirus mediated HSV TK/GCV system combined with radiotherapy on human bladder carcinoma cells. Methods By using retrovirus mediated gene transfer technique,hygromycin phosphotransferase thymidine kinase fusion gene (HyTK) was transferred into EJ bladder carcinoma cells. Antitumor effects were observed after irradiation with 60 Co γ-rays as well as gancyclovir treatment. Results EJ bladder carcinoma cells transduced with HyTK gene and treated with low concentrations of gancyclovir,which alone showed little cytotoxicity,were highly sensitive to radiation. The sensitizer enhancement ratio (SER) was 1.36. Conclusion The combination of retrovirus mediated HyTK/GCV gene therapy with standard radiation therapy might improve the therapeutic effectiveness for bladder carcinoma in clinical practice. |
