Objective To study the changes of expression and function of IL-3R of BMNC and explore the role of IL-3R in the hematopoietic regulation in combined irradiation burn injury in mice. Methods Mice were randomized into normal (N),irradiation (R),burn (B) and combined irradiation burn injury (C) groups. Changes of positive rate,mean contents and function of IL-3R α subunit were determined on the 1st,3rd,7th and 14th days post injury. Relationships between BMNC counts and positive rate,mean contents, function of IL-3R were also evaluated. Results ① Compared with Group N the positive rate and function of IL-3R α subunit of BMNC were significantly decreased in groups R,C,and maintained low during the first 14 days. ② The mean contents of IL-3R α subunit of BMNC were significantly lower in groups R,C than that in group N at first,and then began to increase on the 14th day but did not reach the normal level. There was a significant positive correlation between mean contents of IL-3R α subunit and BMNC counts in all groups. ③ No significant changes in positive rate,mean contents and function of IL- 3R were found in group B. Conclusions ① Expression of IL-3R α of BMNC plays an important role in the damage and recovery of hematopoiesis in combined irradiation burn injury in mice. The funciton of IL-3R of BMNC is also important in the damage of hematopoiesis,but its functional recovery occurs later than that of its content in combined irradiation burn injury in mice. ② There are no significant changes in IL-3R of BMNC after Ⅲ degree burn with 15% TBSA,so combined irradiation burn injury might exert a similar effect on IL-3R as irradiation alone does. |
