Abstract::
      Objective Comparisons of radiosensitivity in cells derived from different individual or pathologic types of nasopharygeal carcinoma were carried out to evaluate the combined detection of radiosensitivity of nasopharygeal carcinoma. Methods The radiosensitivity of nasophargeal carcinoma cell lines CNE and CNE-2Z which had been treated with γ-rays were detected by in situ nick translation(ISNT).The premature chromosome condensation(PCC) technique and cytokinesis-block micronucleus(CBMN) method were used to study radiation-induced cytogenetic damage in cell derived from biopsy samples of 52 patients with nasopharygeal carcinoma. Results The radiosensitivity was lower for CNE which was highly differentiated cancerous cell line than that of CNE-2Z whose differentiation degree was poorer.When the cell lines were exposed to from 1 to 8 Gy of γ-rays, the incorporation rate of dNTP of the two cell lines increased linearly with increasing dosage.The sutdy of PCC assay in biopsy samples from 32 patients with nasopharygeal carcinoma suggested that when the cells were in late G-1 phase and had higher survival rate after irradiation, they were more sensitive than those in other phases.The results of CBMN study in 20 patients also exhibited that the lower micronucleus rate, the easier recurrence of cancer. Conclusion The combined detection of radiosensitivity in cells of nasopharygeal carcinoma may provide potential tool for predicting radiosensitivity of carcerous tissues treated with γ-rays.
