Objective Effects of interleukin-1(IL-1) on proliferation of fibroblasts and their mechanism were analysed in order to provide experimental basis for studying antifibrotic activity from the point of cytokines. Methods MTT colorimetry, a silver colloid techniqiue of AgNOR-S, and immuno-histochemical methods of anti-type Ⅲ collagen and anti-bromodeoxyuridine(BrdUrd) were used to observe the proliferative activity, AgNOR-S and DNA synthesis and expession of type Ⅲ collagen in normal and irradiatd (30 Gy single irradiation by60Co γ-rays) fibroblasts. Results When IL-1 of 100×10+3 U/L was used, the counts and optical density(A) value of normal fibroblasts were 1.4 and 1.3 times higher than those of control group respectively, at the third day after culturing.At the same time, the mean number of AgNOR-S and percentage of normal fibroblasts in S-phase were 2.9 and 2.0 times higher than those of control group, while the number of type Ⅲ collagen positive fibroblats increased by 1.9 times. Conclusion In a definite range of concentration, IL-1 could promote proliferative activity, AgNOR-S and DNA synthesis and expression of type Ⅲ collagen of fibroblasts. |
