Objective To investigate variations in morphology and number of astrocytes in the rat forebrain after the caudate putamen nucleus was irradiated by differant single doses of gamma knife from 10Gy to 100Gy. Methods Immunohistochemical ABC method was used to stain glia fibrillary acidic protein(GFAP). Results Firstly, at dosage of 10-30Gy, the number of astrocytes was increased remarkably in the target area and the non target areas displayed much less reactive cellsand when irradiated with 40-60Gy, the brain showed more hypertrophic and degenerated astrocytes in the target areawhereas at dosage of 70-100Gy, necrotic hole began to appear and became bigger with an increasing dosage.The nucleus caudate putamen and septal area became swollen and blood vessels dilated, which tended to increase with increasing dosage.Lastly, in areas not directly irradiated by gamma knife, the ipsilateral and contralateral sides showed a less difference in GFAP expression in the forebrain. Conclusion Degree of damage in target area is positively relative to the dosage. |
