Objective Application of glycophorin A locus mutation assay as a new biodosimeter. Methods Using flow cytometry and immunolabeling to detect erythrocytes with variant glycoproteins on the surface membrane. Results Among 10 victims of radiation accidents 4 persons were determined as having MN heterozygotes with estimated dose of 1.46-2.9 Gy, and one suspected of having MN. Hemizygote NO variant frequency showed significant doseresponse linear regression with slope of 41.53x10-6/Gy, while NN showed no significant regression. Conclusion Doseresponse data are more fitted to the linearquadratic model than to the linear model. Dose reconstructed with the linearquadratic model is higher than that estimated with the latter. And doses estimated with standard curve of Hiroshima and Chernoby1 are higher than those assessed by assay of dicentrics plus rings after the accidents. |
