Objective Gene expressions of nitric oxide synthase (NOS) and endothelin (ET) as well as ET converting enzyme (ECE) in rat lung after irradiation and their roles in genesis of pulmonary fibrosis were investigated. Methods Rat lung was irradiated with 30Gy of 60Coγ-rays and pulmonary total RNA was extracted at days 4, 8 and 24 after irradiation, respectively.RT PCR was made.Western blot of ET peptide in lung tissues was performed at 1, 3 and 5 months after irradiation.Meanwhile in situ hybridization of NOS mRNA in rat pulmonary tissues was made at 5 months after irradiation. Results NOS mRNA transcription in pulmonary tissues turned into negative;ET mRNA transcription displayed strong positivity; positive ECE mRNA transcription appeared at days 8 and 24 after irradiation and ETB-R mRNA transcription turned to negative at day 24 after irradiation.Western blot of ET in plumonary tissue all displayed positive at 1, 3, and 5 months after irradiation, especially at 1 month.In situ hybridization of NOS mRNA was negative at 5 moths after irradiation. Conclusion Dysregulation of balance between ET and NO may be involved in genesis of radiation pulmonary fibrosis. |
