Abstract::
      Objective To study the neoplastic transformation of human cells in vitro induced by alpha particle irradiation.Methods Neoplastic transformation of SV40 immortalizational human fetal tracheal fitroblast cells was induced by alpha particle irradiation, and changes of cell biological character during the neoplastic transformation were observed. Result The cell morphology was changed after alpha particle irradiation, the relative surviving fraction of the cells irradiated by different doses of alpha particles was dose dependent, and the survival fraction exponential decreased with increasing alpha particles dose. The pattern of cell survival was fitted to single bit single target model. There were aneuploid and instable structure changes in chromosomes, and two marker chromosomes at the same time. The colony forming efficincy in soft agar increased with the passaging. There were 4 tumors after SC injection of the 40th passage cells in 5 nude nice.Conclusion Neoplastic transformation of immortalizational human fetal tracheal fibroblasts can be induced by alpha particles.
