Objective To explore the mechanism of and protective strategy againt bone marrow failure caused by radiation injury.Method Northern blot technique was used to detect the bcl-2, bax and bcl-xL gene expression in murine bone marrow nucleated cells (BMNC) both in Chinese drug treated group which was given per os compound blood activating decoction immediately after irradiation with 8.0 Gy 60 Co γ rays, and in control group.Results On the 3rd and 7th day after irradiation, bcl-2/bax value of BMNC decreased obviously.No bcl-xL expression was observed in control group.In Chinese drug treated group, the bcl-2/bax value increased remarkably, while bcl-xL expression occurred obviously.On the 7th day after irradiation, the bcl-2/bax value in splenic mononuclear cells was consitstent with that in BMNC in both Chinese drug treated and control groups.Conclusion Compound blood activating decoction could have a regulative effect on the expression of cell cycle regulation gene bcl-2, bax and bcl-xL in irradiated bone marrow hematopoietic cells and protect the bone marrow hematopoiesis. |
