Objective To study the effect of rhBMP-2m on irradiated mice.Method 104 male BALB/c mice were randomized to several groups and exposed to 6.5-7.5 Gy whole body 60Co γ rays.After irradiation a dose from 0.5 to 2.0 mg rhBMP-2m in saline were injected i.p. once daily in 1 6 consecutive days, and the animals' 30 day survival was scored.Some indices with relation to hematogenesis were observed as well at ninth day after irradiation.Results It was showed that the 30 day survival, mean survival time, count of bone marrow cells, DNA content of bone marrow cells, count of CFU-GM on agar culture, peripheral blood WBC count, spleen/body weight ratio and number of spleen colony formation were all significantly higher in 2.0 mg rhBMP-2m treated groups than those in the corresponding irradiated controls (P<0.01).Conclusion The beneficial effect of rhBMP-2m on the hemaotogenesis in irradiated mice can be ascribed supposedly to that the rhBMP-2m may induce the release of hematopoietic growth factor(s) in irradiated animal body, or it may act as a hematopoietic growth factor itself, or it may induce a hematopoietic microenvironment to support the growth of stem cells. |
