Objective To study the effect of L-NAME on radiation induced injury of small intestinal epithelial cells 6(IEC-6).Methods Rat IEC-6 cells were irradiated with 60Coγ rays.The content of nitric oxide(NO) in media supernatant of IEC-6 cells was measured by fluorometric method.Nitric oxide synthase (NOS) activity was observed by histochemistry of NADPHdiaphase and immunocytochemistry.Cell viability was evaluated by method of MTT.Results NOgeneration by IEC-6 cells was significantly increased after irradiation.The assay of cNOSactivity showed that it was stimulated.iNOSactivity was inhibited.L-NAME was able to increase cell viability which had been reduced by irradiation, but it was reversed partly by pretreatment with NOsubstrate L Arg.Conclusion NOwas involved in IEC-6 cell injury induced by radiation and L-NAME could provide protective effect on it. |
