Objective To investigate the relationship between mechanism of radiation induced apoptosis and the expresion of p53,bcl-2 genes.Methods Immunohistological staining assay (ABC assay) was used to detect the changes of p53,bcl-2 after radiation.p53 mRNA was examined by in situ hybridization in BT325 cell line after low dose radiation.Results ①Overexpression of p53 was detected in apoptotic groups (vs.control: P <0.01).Expression of bcl-2 decreased in the apoptotic group (P <0.01).The labeling index (LI) of p53 gene was significantly negatively correlated with the LI of bcl-2(0.01< P <0.05),②The LI of p53 mRNA in apoptotic groups was significantly higher than that in the control (P <0.01).Oncoprotein of p53 gene was significantly positively correlated with transcription of p53 mRNA(P <0 01).Conclusion p53 gene has promotive or inhibitory effect on radiation induced apoptosis.bcl-2 gene is an antagonist to apotosis.p53 gene and bcl-2 gene have synergetic or antagonistic relationship in the process of apoptosis. |
