Abstract::
      Objective The aim of this study was to investigate the apoptosis of thymocytes in 60 Coγ irradiated mice.Methods By flow cytometry,electrophoretic analysis of DNA and lymphocyte stimulating test,we determined the changes of thymocyte apoptosis in mice.The mice were whole body irradiated with 2.0,4.0 and 6.0 Gy 60 Coγ rays.Results While the thymocytes underwent apoptosis within 2 h after irradiation,fragmentation rate increased approximately 4~8 h after exposure to radiation doses of 2.0—6.0 Gy.Agarose gel electrophoresis of DNA in the 27 000×g supernatant of lysates obtained from thymocytes of irradiated mice showed ladder pattern.We found that PHA and rIL-2 prevented thymocytes from undergoing apoptosis induced by γ irradiation.Conclusion These observations suggest that the radiosensitivity of mouse thymocytes varied in different dose groups after irradiation.It is possible that PHA and rIL-2 exert protective effects on radiation iuduced apoptosis of thymocytes.
