Objective The HPRT gene mutation spectrum in peripheral lymphocytes in five persons accidentally irradiatied with 2. 0- 5 .2 Gy of 60Co γ-rays was monitored in order to estimate the late effect of radiation. Methods The mutation spectra of whole HPRT gene were analyzed by multiplex polymerase chain reaction using 8 pairs of exon primers and followed by agar gel electrophoresis. Results Exon deletion was the chief feature of HPRT gene mutation induced by ionizing radiation.As the time elapsed, the total percentage of exon deletion decreased gradually from 75% (18/24) at 3 .5 years to 67. 4% (31/46) at 4 5 years, and to 40% (12/30) at 5- 5 years.On the other hand, the total percentage of point mutation elevated stepwise correspondingly.This might reflect the gradual repair of DNA damage in vivo and gradual recovery of the patients. Conclusion Significant changes occurred in the mutation spectrum although HPRT gene mutation remained in the body for quite a long time.Follow up study should be continued. |
